Sean Murphy nasce a Nashua, NH, nell'autunno del 1980. Si è interessato ai fumetti fin dalle elementari e ha trascorso molti pomeriggi della sua infanzia disegnando invece di fare nuove amicizie.
La sua educazione formale è iniziata a Salem, NH come apprendista di Leslie Swank, pittore locale, vignettista e veterano della seconda guerra mondiale. Dopo il diploma di scuola superiore alla Pinkerton Academy nel 1999 ha frequentato il Massachusetts College of Art (Boston) e poi il Savannah College of Art and Design.
Ha iniziato a lavorare professionalmente prima di laurearsi con titoli come Star Wars e Noble Causes, per poi trasferirsi a Hollywood dove ha lavorato su diversi altri progetti per la Dark Horse Comics. La sua carriera ha iniziato a progredire quando la DC Comics gli ha offerto titoli come Batman e Teen Titans. Nel 2005 ha pubblicato il suo primo romanzo grafico originale chiamato Off Road, che ha vinto un American Library Association premio per i giovani adulti. L'anno scorso ha visto l'uscita del suo secondo titolo OGN Orbit esterno (co creato e disegnato con l'amico Zach Howard). Ultimamente ha lavorato con Vertigo Comics su un progetto con Neil Young, freelance con concept art per i vari studios di Hollywood, facendo copertine di Marvel Comics e lavorando al suo prossimo OGN, Punk Rock Gesù.
Attualmente vive a Brooklyn, NY, con la sua fidanzata Colleen e i loro due cani.
